Synopsis

Acute myeloid leukemia (AML) is a malignant disorder of clonal immature white blood cells of the myeloid lineage characterized by the presence of immature blasts (white blood cell precursors) detected in peripheral blood or bone marrow. Blast cells must comprise 20% of bone marrow for a diagnosis of leukemia. Peripheral blood may show abnormalities ranging from pancytopenia to leukocytosis. Most AML cases are spontaneous; however, AML may be a consequence of prior chemotherapy (treatment-related or secondary). AML may arise from underlying myelodysplasia. About 10% of cases are characterized as AML with recurring genetic abnormalities; these abnormalities may influence prognosis and response to chemotherapy. Genetic abnormalities are found by FISH or karyotype, including a specific FLT3 mutation.
